I. Purpose
Genotype, phenotype, and environment form the iron triangle of genetics research. Phenotype (trait) is the result of the combined effects of genotype and environment, with multiple relationships existing between genotype and phenotype. It has become common for researchers to use sequencing and genome resequencing to assess allelic differences and locate quantitative traits, but this requires extensive trait data for validation. However, such analytical measurements are highly susceptible to interference from personnel, tools, and the environment, and can also damage plants. The efficient and accurate Wanshen PhenoGA Plant Phenotype Analysis and Measurement Instrument enables precise data analysis and phenotypic testing with visualization, such as testing phenotypic responses to stress and environmental factors, ecotoxicology tests or germination assays, genetic breeding research, mutant screening, plant morphological modeling, growth studies, etc.
II. Main Performance Indicators
1. Imaging
(1) Dual-color cameras: Top-view and side-view ultra-large zoom lens autofocus Canon EOS DSLR cameras with over 24 megapixels directly connected to a computer to capture top-view and side-view RGB color images of plants.
(2) Infrared binocular 3D camera: An active infrared binocular 3D camera (point cloud density 1024*1024 pixels) at the top captures 2.5D depth pseudo-color images of the plant canopy and 2.5D reconstructed pseudo-color images with adjustable viewing angles for recording and observation.
(3) Imaging chamber: External dimensions 200 cm height * 120 cm length * 120 cm width, capable of imaging and analyzing plants up to 150 cm tall, with a maximum measurable leaf canopy of 115 cm * 115 cm.
2. Analysis Software
(1) Conventional analysis: Projected leaf area and its dynamic changes, outer perimeter, circumscribed circle diameter and area, fitted ellipse major and minor axes and deflection angle, convex hull inner diameter, area and perimeter, plant height and width, minimum bounding rectangle length and width, plant compactness.
(2) Color analysis: RGB, LAB color values, with automatic leaf color correction features, built-in Royal Horticultural Society RHS Color Chart 2015 Edition and Chinese Color System Standard GB/T15608-2006 Color Chart for color comparison. Clustering segmentation based on specified color numbers, and statistics on color distribution and area proportion.
(3) Skeleton analysis: Skeleton length, number of endpoints (number of leaves), number of bifurcations (number of branches, number of nodes), branching angle or stem-leaf angle, etc. Includes the function of connecting endpoints to junctions.
(4) Corn plant shape analysis: Number of leaves, leaf length and width, leaf curvature, leaf canopy projected area, number of stem nodes, node length and thickness, leaf color, etc., with editable features.
(5) Growth analysis: Absolute growth and relative growth curves of plants, relative growth trends.
(6) Root analysis: Root length, root thickness, number of root tips, etc. (requires root thickness >1mm).
(7) Seed analysis: Number of seeds, seed area, seed length and width (seed diameter >2mm, non-adhesive), analysis of seed morphology, fruit appearance quality, flower shape (compactness, bounding rectangle length and width, and other shape parameters), and flower color.
(8) Others: Automatic batch processing and analysis at different growth stages, multi-plant grid analysis, geometric measurements such as lines and angles, editable and correctable measurement results.
3. Data Reporting
(1) Can connect to a barcode scanner to automatically input sample numbers, with features for tracking analysis based on barcode identification.
(2) All analysis data and marked images can be exported.
